Synthesis and conformational studies of a stable peptidomimetic -hairpin based on a bifunctional diketopiperazine turn inducer

چکیده

The design, synthesis and conformational studies of a new b-hairpin mimic, 2, are described in this paper. The design towards hairpin 2 is based on the assembly of the bifunctional DKP-1 as a b-turn inducer, a peptidomimetic strand, namely 5-amino-2-methoxybenzhydrazide, to stabilize the formation of b-sheets, and finally a tetrapeptide sequence, GVVI, containing the hydrophobic residues Val, Ile and Gly. The synthesis of hairpin 2 was performed in solution while the formation of the b-hairpin in protic solvents was proven by NMR investigations (H and C chemical shifts, vicinal coupling constants and ROEs) and corroborated by computational studies (Monte-Carlo conformational search, molecular dynamics and DFT calculations).

β-Hairpin stabilization through an interstrand triazole bridge.

β-Hairpin peptides were conformationally stabilized through a 1,4 disubstituted 1,2,3-triazole interstrand linkage. A NMR conformational analysis revealed that the β-hairpin content depends on the number and position of substituent methylene units of the 1,2,3-triazole ring. A simple method for the synthesis of a sugar furanoid trans vicinal diacid and its incorporation into the N-terminal tetrapeptide sequence (H-Phe-Trp-Lys-Thr-OH) to get glycopeptide has been described. 2D NMR and MD simulation studies of clearly show that the sugar diacid adopts a γ-turn conformation towards the N-terminus.
